Effects of Medium Composition on Mass - Production of the Multiple Shoots from a Shoot in Lilium Oriental Hybrid ¢¥ Casa Blanca ¢¥ In Vitro

Abstract
To enhance mass-production of multiple shoots from Lilium Oriental Hybrid ¢¥Casa Blanca¢¥, several factors such as light, levels of total inorganic salt, agar, growth regulators, and vitamins on MS medium were examined in vitro. The number and growth of shoots were most effective when the plants were exposed to 16 hours of light and cultured for 8 weeks on double strength of basal MS medium. Agar concentration of 6.0 g¡¤L^(-1) was also effective in mass-production of multiple shoots, while shoot growth was more evident without agar on MS medium in liquid shaking culture. More number of shoots increased when MS medium containing 0.1 §·¡¤L^(-1) NAA. The maximum shoot growth was achieved on 0.1 §·¡¤L^(-1) NAA and 1.0 §·¡¤L^(-1) TDZ containing medium under 16 hours of light. A high concentration of thiamin (1.0 §·¡¤L^(-1)) was favorable multiple shoots and pyridoxine requirement was relatively low (0.01 §·¡¤L^(-1)).
